We’ll work closely with you to design suitable assays for your program that will measure key parameters and provide flexibility to maximize the potential for success.

IN PARTICULAR, WE’LL CONSIDER:

For biochemical assays we will ensure:

  • enzyme/protein purity and identity to avoid picking up nonspecific activities
  • buffer optimization, including use of detergents, carrier proteins, chelators
  • enzyme kinetics to ensure robust evaluation and comparison of compound activities (eg Km determination ; KD determination for binding)
  • pre-incubation between enzyme and compounds to allow the detection of slow-binding inhibitors

For cell-based assays, we will:

  • Select appropriate cell model to ensure relevance for the target/phenotype under consideration
  • Identify relevant cell line to generate stable cell lines with reporter or split reporter systems, or to monitor pathway activation and/ or second messenger modulation
  • Define cell treatment time based on the target biology

We apply specific and stringent criteria to qualify, validate, and monitor assays over time and use these validated assays to identify new chemical matter, screening our collection, and to support SAR work.

We recognize the importance of aligning assay quality with throughput rates and apply our extensive experience in high-throughput screening to achieve this. We can also miniaturize assays and will maximize metrics to achieve optimal assay formats.
