About this poster
In this poster, we outline the development of a novel inducible cellular model to support SARS-CoV-2 replication in a safe and controlled manner. Given the virus’s cytopathic nature and BSL-3 classification, we implemented a Tet-Off system to enable conditional expression of viral components, thereby avoiding cytotoxicity and allowing for stable replicon expression. Additionally, we employed an EBV-based episomal vector to address challenges posed by the large size of the SARS-CoV-2 genome, enabling stable maintenance of the replicon without genomic integration. This system lays the foundation for high-throughput antiviral screening in a BSL-2 setting, facilitating the identification of candidate molecules targeting SARS-CoV-2 replication.
